Standard Operating Procedure for UV-Vis Spectrophotometers

1. Purpose In order to make the instrument work normally and ensure the reliability of the results, this operating procedure is specially formulated. 2. Scope of application Laboratory 3. Responsible The laboratory personnel should strictly follow the operating procedures, and the QC supervisor is responsible for supervising the implementation of the procedures. 4. Definitions None 5. Safety Precautions If an organic solvent is used as a solvent, it should be noted that it is harmful to human health. 6. Procedure 6.1. Preparation 6.1.1 Power on: Turn on Sipper (suction machine), 8453 detector, monitor, computer host and printer power in turn, and allow the instrument to warm up for 30-45 minutes. 6.1.2 Select Windows NT Version 4.00. 6.1.3 Press"Alt + Ctrl + Del" key to log in. 6.1.4 Enter User Name in turn:"Administrator", and PassWord:"3000hanover" Enter the Window-NT main screen. 6.1.5 Press"START" key, use the mouse to click Program, and then click HP-UV-Chemstation to enter the workstation state, select"Instrument on line". 6.2. Test operation process 6.2.1 Tap dialog box"Mode" key to select from its menu"Standard" mode, by"Instrument" Under the item, select lamp to turn on the deuterium lamp or tungsten lamp. 6.2.2 Determination of absorbance at a fixed wavelength·Tap the Task button and select from the menu"Fixed Wavelength".​​·Tap the Set Up key next to the Fixed WL key to set the following measurement parameters: Enter the set wavelength. Enter the background correction method to select the data mode.​​·tap"Sampling" dialog, in its menu select"Sipper".​​·Will "Sipper" Immerse the pipette in the solvent used to prepare the test solution, tap lightly"Blank", to do a blank scan.​​·Will "Sipper" Dip the pipette into the sample solution and tap"Sample" Scan the test solution and measure the absorbance value of the sample at this wavelength. 6.2.3 Full-wavelength scanning·tap"Task" box, select from the menu"spectrum/peaks".​​·tap"spectrum/peaks" right side "Set Up" key to set the following parameters: the number of absorption peaks and valleys the wavelength range of the scan to enter the desired data model (Data Mode)·tap"Sampling" dialog, in its menu select"Sipper".​​·Will"Sipper"Immerse the pipette in the solvent used to prepare the test solution, tap lightly"Blank" Do a blank scan.​​·Will "Sipper" Dip the pipette into the test solution and tap"Sample" Scan the test solution and determine the absorbance value of the test solution. 6.2.4 Content determination by working curve method·tap"Task" dialog, select"Quantities".​​·tap next to"Set Up", set the following parameters. Enter the measurement wavelength. Select the calibration curve. Select the Data Mode. Enter the appropriate unit.​​·tap"Sampling" dialog, in its menu select"Sipper".​​·Will“Sipper"Immerse the pipette in the solvent used to prepare the test solution, tap lightly"Blank", to do a blank scan.​​·tap“StandardTable", Under this screen, measure the reference solution of different concentrations, and obtain the corresponding standard curve, enter the name of the reference solution and its corresponding concentration in the reference product column.​​·Will "Sipper" Dip the pipette into the test solution, tap the bottom left"Sample", scan the test solution and measure the absorbance of the test solution at this wavelength.​​·tap"SampleTable", enter the name of the test solution and the dilution factor in the dialog box it opens, and read the measured content. 6.3. End of work 6.3.1. Cleaning of Sipper pipeline: first clean the pipeline with the solvent used for the sample to be tested, and then clean it with pure water. 6.3.2. Print scan map and test data. 6.3.3. Exit UV-Chemistation, exit Windows-NT. 6.3.4. Shut down in the reverse order of startup and cut off the power. 6.3.5. Fill in the usage record.
